Non-volatile reaction mixture의 qualitative examination에 사용이 되는 simple하고 rapid한 일종의 liquid chromatograghy로 glass, aluminum 또는 plastic plate에 solid stationary phase가 coating되어있다.

(1) 사용되는 경우
- Sample purification monitoring

(2) Adsorbent (Stationary phases)
- Silica gel, alumina, kieselguhr, cellulose powder
- Fluorescent compound에 사용이 가능하며 mixture compound의 resolution은 ultraviolet light하에서 확인이 가능하다.
- Adsorbent는 hydrated calcium sulfate와 같은 binder와 함께 혼합되어 supporter(glass plate or plastic sheet)에 layer를
고착시킨다.
- 가장 널리 사용하는 stationary phase는 silica gel과 alumina이다.
(3) Preparation of plates
- 요즘은 바로 사용하도록 상업화된 TLC plate를 구입하므로 plate를 특별히 제작할 필요는 없다.
(4) Two-dimensional Chromatogram
- 1-D TLC로 분리가 잘 되지 않는 것을 larger plate를 사용해 두 방향으로 TLC를
올려서 분리가 잘되게 하는 것이다.
- 두 방향으로 올려주면 solvent의 travel distance가 길어져서 분리가 더 잘된다.
- 각 방향에 대해서 solvent system을 다리게 사용해야 한다.
- 첫 번째 solvent system으로 elution을 해 준 다음, TLC plate를 90° 돌려서
두 번째 solvent system으로 elution을 해준다.
- UV로 관찰할 수 있는 samples만 가능하다.(발색할 수 없음.)
- 1-D에 비해 효용성이 떨어진다.
